Target Information
The 26S proteasome is a multicatalytic proteinase complex composed of a 20S core and a 19S regulator. The 20S core contains four rings of 28 non-identical subunits (two alpha and two beta rings). The 19S regulator includes a base with six ATPase and two non-ATPase subunits, and a lid containing up to ten non-ATPase subunits.
Proteasomes degrade peptides in an ATP/ubiquitin-dependent, non-lysosomal pathway and play a key role in MHC class I peptide processing.
The PSMC4 gene encodes one of the ATPase subunits of the 19S regulator, belonging to the triple-A family of ATPases with chaperone-like activity. This subunit interacts with nuclear hormone receptor family members and the liver oncoprotein gankyrin. Two transcript variants encoding different isoforms have been identified.
