Target Information

Influenza A H1N1 NS, also known as Non-structural protein 1 (NS1A or NS1), is a 234-amino-acid protein from Influenza A virus (strain A/Swine/Iowa/15/1930 H1N1), which infects birds, humans, and pigs.
It belongs to the NS1 family of influenza A viruses and exists as a homodimer located in the nucleus of uninfected or transfected cells, and in the cytoplasm of infected cells.

The viral protein NS1 binds and inhibits two cellular proteins required for 3'-end processing of cellular pre-mRNAs — CPSF4 (30 kDa cleavage and polyadenylation specificity factor) and PABPN1 (poly(A)-binding protein 2).
This inhibition blocks post-transcriptional processing of cellular pre-mRNA, causing accumulation of unprocessed pre-mRNAs in the nucleus and loss of cellular protein synthesis.
As a result, the virus evades interferon synthesis after activation of the IRF3 pathway, preventing the establishment of an antiviral state.
Additionally, NS1 prevents activation of human EIF2AK2/PKR early in infection, further suppressing the antiviral response.
