Target Information
두 개의 유전자가 type II integral membrane protein 계열의 새로운 구성원을 암호화합니다. 이들은 광범위하게 발현되며, 조직 특이적인 mRNA 개시 및 스플라이싱을 통해 주요한 두 가지 isoform을 생성합니다. 작은 isoform은 N-말단이 절단된 형태입니다. 이러한 단백질들은 nuclear envelope spectrin repeat의 존재로 인해 nesprin-1 및 -2로 명명되며, bipartite nuclear localization sequence와 보존된 C-말단 단일 transmembrane domain을 포함합니다. EGFP-fusion 발현 실험에서 핵막에 위치함이 확인되었으며, perinuclear localization에는 C-말단 TM-domain이 필수적입니다. Nesprin-1은 평활근 및 골격근에서 발달적으로 조절되며, myoblast 분화 동안 핵막에서 핵 및 세포질로 재배치됩니다. Nesprin 단백질은 핵의 구조적 안정성과 조직 유지를 위한 “dystrophins of the nucleus” 역할을 할 수 있습니다.
