Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to signaling pathways, regulating cell death and stress responses.
TRAF proteins share a homology region enabling binding to receptors and other TRAFs, activating distinct signaling cascades.
TRAF2 and TRAF3 bind directly to CD40, a member of the NF receptor superfamily, and are critical for NF-kappa-B activation in B lymphocytes.
TRAF2, along with TRAF6, is essential for CD40 signaling in non-hematopoietic cells and interacts with the lymphotoxin-beta receptor (LTbetaR) in association with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
