Product Specific Information

The sym0F1 monoclonal antibody reacts with human and mouse c-Maf, a 42 kDa basic leucine zipper transcription factor involved in neural, ocular, and hematopoietic systems.
In hematopoietic cells, c-Maf is crucial for IL-4 expression in Th2 cells and is also important for Th17 and Tfh cell differentiation and function, driving IL-21 expression and regulating IL-23R and CXCR5 expression.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested for intracellular staining of Th17-polarized mouse splenocytes using the Foxp3/Transcription Factor Buffer Set and protocol.
Use at 5 µL (0.0075 µg) per test in a final volume of 100 µL (cell number 10⁵–10⁸ cells/test).

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm).
Recommended filter: 710/50 bandpass (alternative: 695/40 bandpass).

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark.

Excitation/Emission: 488 nm / 710 nm (Blue Laser)
Filtration: 0.2 µm post-manufacturing filtered


Target Information

c-Maf is the cellular counterpart of oncogenic v-Maf, a basic leucine zipper transcription factor.
It regulates IL-4 expression in Th2 cells, crystalline genes in lens fiber cells, insulin gene in islets, and p53/L7 genes through binding to Maf recognition element (MARE).
c-Maf also regulates Th2 differentiation and IL-10 expression in macrophages.
Chromosomal aberrations involving maf are associated with multiple myeloma, where it is expressed in myeloma cell lines and resting monocytes/macrophages.
