Target Information

FOXO1 (FKHR, Forkhead Box O1) is a member of the forkhead homeotic gene family of transcription factors.
FOXO1 can act as a coactivator or corepressor of nuclear receptor activity through the LXXLL motif in its C-terminal region.
It is implicated in alveolar rhabdomyosarcoma via association with PAX3 and plays roles in autophagy, myogenic differentiation, and tumor suppression.
FOXO1 is a primary target of insulin signaling, regulating metabolic homeostasis under oxidative stress.
It binds to insulin response elements (IRE) and Daf-16 family binding elements (DBE), influencing redox balance, osteoblast regulation, bone mass control, and glucose metabolism.
In hepatocytes, FOXO1 promotes gluconeogenesis by activating genes such as IGFBP1, G6PC, and PPCK1 in cooperation with PPARGC1A.
It acts downstream of CDK1, PKB/AKT1, and SKT4/MST1 in regulating cell death.
