Target Information
L-fucose is a key sugar component in glycoproteins and other complex carbohydrates, playing roles in cellular recognition and signaling. The fucosyl donor for fucosylated oligosaccharides is GDP-beta-L-fucose. There are two pathways for GDP-fucose biosynthesis:
- The major pathway converts GDP-alpha-D-mannose to GDP-beta-L-fucose.
- The alternate pathway, active in mammalian tissues such as liver and kidney, functions as a salvage route to reutilize L-fucose from glycoprotein and glycolipid turnover.
In this pathway, L-fucose is phosphorylated to beta-L-fucose-1-phosphate, which then condenses with GTP via fucose-1-phosphate guanylyltransferase to form GDP-beta-L-fucose.
