Target Information

EIF2AK2 (PKR) is one of four kinases that specifically phosphorylate Ser51 of translation initiation factor eIF2-alpha in response to environmental stresses. It plays a crucial role in antiviral defense through activation by double-stranded RNA produced during viral infection. PKR mediates IFN-induced innate antiviral responses, virus-induced apoptosis, and regulates cell growth and differentiation. It can induce apoptosis via Fas up-regulation and FADD/Caspase-8 signaling. PKR also modulates multiple pathways including NF-κB activation, p53, p38, and PDGF signaling. It has been implicated in tumor suppression, malignancy, and neurodegenerative diseases such as Alzheimer’s, Huntington’s, and ALS. Viruses often inhibit PKR to evade host defenses, such as through influenza virus NS1 protein.
