Product Specific Information

The NP-6G4 monoclonal antibody reacts with human CD195 (CCR5), an approximately 45 kDa member of the G protein-coupled receptor (GPCR) superfamily that binds chemokines including MIP-1α, MIP-1β, and RANTES. CCR5 is expressed on resting T cells with a memory/effector phenotype, monocytes, macrophages, and immature dendritic cells. It functions as a receptor directing lymphocyte chemotaxis and serves as a primary co-receptor for macrophage-tropic HIV-1.

• Applications Tested: Pre-titrated and tested on normal human peripheral blood cells
  • Recommended staining: 37°C for 1 hour in Flow Cytometry Staining Buffer (Product #00-4222-57)
  • Use at 5 µL (0.125 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test)

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). Use a 710/50 bandpass filter (695/40 acceptable alternative). Ensure instrument compatibility.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal brightness loss. Clone-specific performance should be determined empirically.

Target Information

Human immunodeficiency virus (HIV) and related viruses require coreceptors, in addition to CD4, to infect target cells. CCR5 (CC-CKR-5) is a principal coreceptor for macrophage- and dual-tropic HIV-1 strains, facilitating fusion and entry into white blood cells. CCR5 is essential for infection by HIV-1, HIV-2, and SIV. Its ligands (RANTES, MIP-α, MIP-β) can prevent M-tropic HIV-1 infection. CCR5 mRNA is expressed in lymphoid organs and monocytes.
