This Mags.21F7 monoclonal antibody reacts with human and mouse promyelocytic leukemia zinc finger (PLZF), a member of the BTB-POZ family of transcription factors. Expression of PLZF differs between species:

• Mouse: Highly expressed in immature CD1d-restricted NKT2, NKT1, and subsets of γδ (Vγ1.1+Vδ6.3+) T cells.
• Human: Expressed in NK cells, γδ T cells, CD4+ and CD8+ T cells, MR1-specific mucosal-associated invariant T cells, and MHC Class II-restricted T cells.

PLZF functions as a transcriptional repressor involved in NKT cell development, NK cell function, cellular quiescence, and growth suppression. It also inhibits gene expression induced by retinoic acid receptor.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Intracellular staining of mouse thymocytes at ≤1 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
Careful titration recommended for optimal assay performance.

Target Information

The PLZF gene encodes a zinc finger transcription factor with nine Kruppel-type zinc finger domains located at the carboxyl terminus. This nuclear protein is involved in cell cycle progression and interacts with histone deacetylase. Aberrant rearrangements of this locus are associated with acute promyelocytic leukemia (APL). Alternate splice variants have been characterized.
