Background

  • Domain: PH domain binds phospholipids including phosphatidic acid, PI3P, PI(3,5)P₂, and PI(3,4,5)P₃.
  • Enzyme Regulation: GAP activity stimulated by PI(4,5)P₂ and phosphatidic acid.
  • Function: Acts as a GTPase-activating protein (GAP) for ARF6, essential for clathrin-dependent export of proteins from recycling endosomes to the trans-Golgi network and cell surface.
  • Miscellaneous:
    • Overexpression causes accumulation of ITGB1 in recycling endosomes and inhibits stimulation-dependent cell migration.
    • Reduced ACAP1 or AKT1/2 levels inhibit cell migration.
    • Co-expression with PIP5K1C induces tubular endosomal membrane structures.
  • PTM: Phosphorylation at Ser-554 by PKB is required for ITGB1 interaction and export from recycling endosomes to the cell surface.
  • Similarity: Contains 1 Arf-GAP domain, 1 BAR domain, 1 PH domain, and 3 ANK repeats.
  • Subunit: Interacts with GTP-bound ARF6, SLC2A4/GLUT4, CLTC, and GULP1. Forms a complex with GDP-bound ARF6 and GULP1.
  • Tissue Specificity: Highest expression in lung and spleen; low in heart, kidney, liver, and pancreas.
