Product Specific Information

The 17B5 monoclonal antibody reacts with mouse CD137 (4-1BB), a TNFR superfamily member. CD137 is expressed as monomers, dimers, and tetramers on activated T cells around day 4–6 after activation via the TCR complex. Interaction with its ligand 4-1BBL delivers a costimulatory signal crucial for T-APC interaction. Recent data suggest CD137 is expressed on Tregs and may influence their proliferation. The 17B5 antibody exhibits strong antagonistic effects in in vitro functional assays.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on activated mouse splenocyte cultures at ≤0.25 µg/test (100 µL final volume). Cell count may vary (10⁵–10⁸ cells/test). Titrate for optimal performance.

Target Information

CD137 (TNFRSF9, 4-1BB) is an inducible costimulatory molecule mainly expressed on activated T cells. Its ligand, 4-1BBL, is found on activated macrophages, mature B cells, hematopoietic stem cells, and myeloid progenitors. CD137 signaling promotes T cell survival, memory CD8⁺ T cell maintenance, clonal expansion, and regulates cytokine release affecting monocyte survival. Soluble CD137 may act as a negative regulator in immune responses.
