Background
The protein encoded by the RUBIC gene is a negative regulator of autophagy and endocytic trafficking, controlling endosome maturation. It contains two conserved domains: an N-terminal RUN domain involved in Ras-like GTPase signaling, and a C-terminal DUF4206 domain with a diacylglycerol (DAG) binding-like motif. Mutation in this gene can cause recessive ataxia due to deletion of the DAG binding-like motif. Alternatively spliced transcript variants encoding distinct isoforms have been identified.
