Target Information
CHMP3는 endosomal sorting required for transport complex III (ESCRT-III)의 핵심 구성요소로서, multivesicular bodies (MVBs) 형성과 endosomal cargo 단백질의 분류에 관여합니다.
MVB는 내강 소포(ILV)를 포함하며, 이는 endosome의 제한막으로부터 내 invagination 및 절단 과정을 통해 생성되어 lysosome으로 전달되어 막 단백질, 성장인자 수용체, 효소 및 지질의 분해를 가능하게 합니다.
ESCRT-III 단백질은 ILV 방출 전에 막에서 분리되며, cytokinesis의 최종 단계 및 바이러스 budding(HIV-1 등)과 같은 막 절단 과정에서도 기능합니다.
CHMP3는 phosphatidylinositol 3,5-bisphosphate PtdIns(3,5)P2와 PtdIns(3,4)P2에 선택적으로 결합하며, endosomal sorting 및 EGF receptor trafficking, cytokinesis의 후기 단계에 관여합니다. Isoform 2는 세포 스트레스에 의한 사멸 및 ROS 축적을 억제하는 역할을 합니다.
