F-Box and WD Repeat Domain Containing 7 (FBXW7)는 약 40개의 아미노산 모티프(F-box)를 특징으로 하는 F-box 단백질 패밀리의 구성원입니다. F-box 단백질은 SKP1-cullin-F-box(SCF) 복합체의 4개 서브유닛 중 하나로, 인산화 의존적 유비퀴틴화 과정에서 기능합니다.
F-box 단백질은 구조에 따라 3가지 클래스로 나뉩니다:
- Fbws: WD-40 도메인을 포함
- Fbls: leucine-rich 반복 서열 포함
- Fbxs: 기타 단백질-단백질 상호작용 모듈 또는 인식 가능한 모티프 없음
FBXW7은 Fbws 클래스에 속하며, F-box 외에도 7개의 연속된 WD40 반복을 포함합니다. 이 단백질은 cyclin E에 직접 결합하며, cyclin E의 유비퀴틴 매개 분해를 유도하는 것으로 알려져 있습니다.
이 유전자의 돌연변이는 난소암 및 유방암 세포주에서 발견되어, 인간 암의 발병 기전에 관여할 가능성이 제시되었습니다. 다양한 전사체 변이가 보고되어 서로 다른 isoform을 암호화합니다.
