Intercellular Adhesion Molecule-3 (ICAM-3) is a member of the immunoglobulin supergene family and functions as a ligand for Lymphocyte Function-Associated Antigen-1 (LFA-1). ICAM-3 is a heavily glycosylated, integral membrane protein with five immunoglobulin-like domains. It shares high homology with ICAM-1 and ICAM-2 in the extracellular region but is absent on endothelia. ICAM-3 is expressed on resting lymphocytes, monocytes, and neutrophils, representing the major LFA-1 ligand on these cells. The adhesion of resting T lymphocytes to LFA-1 primarily via ICAM-3, along with its high expression on monocytes and resting lymphocytes, suggests an important role in initiating immune responses.

Target Information

CD50 (Intracellular Adhesion Molecule 3, ICAM-3) is a transmembrane glycoprotein expressed by leukocytes and serves as a counter-receptor for the LFA-1 integrin. It mediates contact between T cells and antigen-presenting cells, regulates LFA-1 affinity for ICAM-1, and induces T cell activation and proliferation. ICAM-3 plays an essential role in initiating immune responses on both T cells and antigen-presenting cells. It also interacts with CD209 (DC-SIGN), a C-type lectin of dendritic cells and macrophages, facilitating communication between dendritic cells and granulocytes.
