Target Information
APOBEC (apolipoprotein B mRNA editing enzyme, catalytic) proteins inhibit retroviruses by deaminating cytosine residues of viral RNA and DNA. The seven APOBEC3 genes or pseudogenes are located in a cluster on chromosome 22, likely resulting from gene duplication.
Like APOBEC3G, APOBEC3F deaminates deoxycytosine to deoxyuracil in the minus strand of HIV-1 DNA, causing G-to-A hypermutation in the plus strand. APOBEC3G and APOBEC3F thus contribute to innate immunity against retroviruses and genetic variation in many viruses.
HIV-1 Vif protein counteracts APOBEC3G and APOBEC3F by impairing translation and promoting proteasomal degradation. Interestingly, mouse APOBEC3G/F resist HIV-1 Vif, effectively inhibiting wild-type HIV-1 replication, whereas human APOBEC3G/F inhibit only Vif-deficient HIV-1. Modulating APOBEC3G/F activity or blocking their interaction with Vif could have therapeutic potential.
