Target Information
Retinoic acid-inducible gene I (RIG-I, DDX58)는 바이러스 dsRNA를 인식하는 패턴 인식 수용체(PRR)입니다. RIG-I와 MDA5는 바이러스성 dsRNA를 감지하여 선천 면역 반응을 활성화합니다. 두 단백질은 RNA helicase로서 중복되면서도 상이한 역할을 수행합니다.
RIG-I는 5'-triphosphate 말단이 없는 짧은 dsRNA에 의해 활성화되며, MDA5는 긴 dsRNA에 반응합니다. 활성화 후 IPS-1을 통해 NF-κB와 IRF-3 전사 인자를 자극하여 세포자멸사, 사이토카인 신호전달, 염증 반응을 유도합니다.
RIG-I는 인플루엔자 A/B, RSV, 파라믹소바이러스, 일본뇌염바이러스, 웨스트나일바이러스 등의 신호전달에 필수적입니다. 또한 microRNA-146a는 RIG-I 의존적 항바이러스 반응을 TRAF6, IRAK1, IRAK2의 음성 조절을 통해 억제하는 것으로 알려져 있습니다. 최근 연구에서는 RIG-I가 RNA polymerase III를 통해 세포질 DNA를 감지할 수 있음이 보고되었습니다.
