Target Information
Furin is a membrane-associated, calcium-dependent serine protease belonging to the subtilisin-like prohormone convertase (PC) family. These enzymes cleave most prohormones and neuropeptide precursors, as well as various cellular, viral, and bacterial proteins transported via the constitutive secretory pathway.
Furin and other PCs share structural features including:
- An N-terminal proregion (~10 kDa)
- A conserved catalytic domain (~55 kDa)
- A variable C-terminal domain
Furin becomes catalytically active after proregion cleavage and is the only PC with a transmembrane domain. Cleavage occurs at the carboxyl terminus of the consensus sequence RX(K/R)R. The acidic sequence C771PSDSEEDEG780 localizes furin to the trans-Golgi network, and phosphorylation within this region affects intracellular routing. The Y759KGL762 motif mediates internalization from the cell surface.
