Product Specific Information
Recombinant antibodies are produced using genes encoding the desired antibody, cloned into an expression vector and expressed in vitro. These antibodies offer improved specificity and lot-to-lot consistency.
Applications Tested:
The CR55T33 clone has been tested for flow cytometry at 0.5 µg per test. A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be empirically determined (typically 10⁵–10⁸ cells/test). It is recommended to titrate the antibody for optimal performance.
Target Information
Histone H2A.X (H2AX) is a core histone protein involved in nucleosome formation. DNA double-strand breaks trigger phosphorylation of serine 139, forming γ-H2AX, which can be detected by Western blot or immunofluorescence to assess DNA damage frequency.
Phosphorylation is mediated by phosphatidylinositol 3-kinases, including ATM, DNA-PK, and ATR. H2A.X consists of 143 amino acids and is synthesized during G1 and S-phase. Its mRNA exists in two forms: one polyadenylated (~1600 bases) and one non-polyadenylated (~575 bases). The gene maps to chromosome 11q23.2-q23.3 and plays a key role in DNA structure and repair by regulating MDC1 binding at damage sites.
