Target Information
CXCR4 (also known as Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor with seven transmembrane domains, functioning as a co-receptor for X4 HIV-1 entry into CD4+ cells. It also serves as a co-receptor for HIV-2 binding to CD4 via gp120 and promotes Env-mediated viral fusion. CXCR4 acts as a receptor for the C-X-C chemokine CXCL12/SDF-1, mediating intracellular calcium increase and MAPK1/MAPK3 activation. It is involved in hematopoiesis, cardiac ventricular septum formation, and vascularization of the gastrointestinal tract. In the CNS, CXCR4 mediates hippocampal neuron survival and cerebellar development. CXCR4 also binds bacterial LPS, mediating LPS-induced inflammatory responses including TNF secretion by monocytes. Antibodies against CXCR4 can block HIV-1 and HIV-2 fusion and infection of human target cells. The amino-terminal domain and the second extracellular loop of CXCR4 serve as HIV binding sites.
