Target Information

Retinoic acid-inducible gene I (RIG-I) is a pattern recognition receptor (PRR) involved in the detection of viral double-stranded RNA (dsRNA). Along with MDA5, RIG-I activates innate immune responses by recognizing viral RNA.

  • RIG-I detects short dsRNAs and those lacking a 5'-triphosphate end.
  • MDA5 detects long dsRNAs.
    Once activated, both signal through IPS-1 to activate NF-κB and IRF-3, leading to apoptosis, cytokine signaling, and inflammation.
    RIG-I is essential for responses to influenza A/B, respiratory syncytial virus, paramyxoviruses, Japanese encephalitis virus, and West Nile virus.
    MicroRNA-146a negatively regulates RIG-I-dependent antiviral responses by targeting TRAF6, IRAK1, and IRAK2.
    Recent studies suggest RIG-I also detects cytosolic DNA via RNA polymerase III activity.
