Target Information
The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with the establishment and maintenance of pregnancy.
PR exists as two forms:
- Form A (94 kDa)
- Form B (120 kDa)
These forms arise from alternative translation initiation sites. PR-B is the transcriptionally active form responsible for activating genes related to endometrial maintenance, pregnancy support, and ovulation inhibition. PR-A is identical to PR-B except for a 165 amino acid deletion at the N-terminus, which exposes a 140 amino acid inhibitory domain (ID) acting as a repressor of steroid hormone transcriptional activity.
In its inactive state, PgR forms a multiprotein complex including heat shock proteins and immunophilins. Upon progesterone binding, a conformational change induces receptor dimerization and binding to progesterone response elements (PRE), leading to transcriptional activation. A null mutation in the PGR gene results in pleiotropic reproductive abnormalities.
