Target Information

Immunophilins are soluble cytosolic receptors that bind immunosuppressant agents such as cyclosporin A (CsA) or FK506.
Proteins binding FK506 are termed FK506 Binding Proteins (FKBPs), and those binding cyclosporin A are cyclophilins (CyP).

Both CyP:CsA and FKBP:FK506 complexes inhibit calcineurin, a calcium/calmodulin-dependent phosphatase involved in T-cell activation, thus mediating immunosuppressive effects.

Immunophilins act as peptidyl prolyl cis-trans isomerases (PPIase) that accelerate protein folding by catalyzing proline isomerization. Within the cyclophilin family, several homologous proteins exist, including CyPA, CyPB, and CyPC.

CyPA (Cyclophilin A), also known as CyP-18, is the most abundant and ubiquitous cyclophilin found in all vertebrate tissues, including T-cells.
