Product Specific Information

This CHI2S4N monoclonal antibody recognizes human and mouse STAT6 phosphorylated on tyrosine 641. Following phosphorylation by JAKs, STAT proteins translocate to the nucleus to regulate gene transcription. In response to IL-4, STAT6 is phosphorylated by JAK1 and JAK3, promoting T cell growth and B cell IgE production.

Specificity was verified by ELISA and flow cytometry.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated for intracellular staining of stimulated normal human peripheral blood cells. Recommended usage: 5 µL (0.125 µg)/test in 100 µL total volume.

Staining Protocol: Use Protocol C (Two-step: Fixation/Methanol). Protocols for intracellular staining are available at Thermo Fisher BestProtocols® online resource.

PE-eFluor® 610 can be excited with 488–561 nm lasers and emits at 607 nm. Recommended filter: 610/20 band pass (PE-Texas Red® equivalent). Ensure instrument compatibility.

Light Sensitivity: Protect vial and samples from light due to photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer or 1-step Fix/Lyse Solution for up to 3 days at 4°C in the dark with minimal brightness loss.
Excitation / Emission / Laser: 488–561 nm / 607 nm / Blue, Green, Yellow-Green lasers.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

STAT6 is a transcription factor activated by cytokines such as IL-4 and IL-13. STAT6-deficient models show impaired Th2 differentiation, IgE class switching, and IL-4-mediated proliferation. STAT6 mRNA is expressed in lymphoid and various tissue types. STAT6 plays a critical role in Th2 immune response and IL-4-mediated biological processes, existing in two isoforms (STAT6a, STAT6c).
