Product Specific Information
The FJK-16s antibody reacts with mouse, rat, dog, porcine, bovine, and cat Foxp3 (FORKHEAD BOX P3, SCURFIN, JM2). Foxp3 is a 49–55 kDa transcriptional regulator involved in immune homeostasis.
Immunoblotting mapped the epitope to amino acids 75–125 of mouse Foxp3. In humans, this region is alternatively spliced, with both isoforms expressed in CD4+CD25+ lymphocytes. Mouse splenocytes lack the alternatively spliced isoform, indicating species-specific regulation.
FJK-16s is optimized for use with the Foxp3/Transcription Factor Buffer Staining Set (Product #00-5523-00). Use of other buffers is not recommended.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested using mouse splenocytes and the recommended staining buffer set. Use ≤0.5 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test (titrate for optimal performance).
Alexa Fluor 660 emits at 690 nm, suitable for spectral cytometers and multiplexing with APC, Alexa Fluor 647, and Alexa Fluor 700.

Target Information
FOXP3 (Forkhead box protein 3) is a transcription factor essential for immune regulation. It is highly expressed in CD25+CD4+ regulatory T cells and absent in CD8+ T cells. FOXP3 mutations cause scurfy phenotype in mice and IPEX syndrome in humans. Multiple isoforms are known, and expression is regulated by p53 in response to DNA damage in certain cancer cells.
