Guanylate cyclase-activating protein (GCAP1) is a Ca²⁺-binding protein that upregulates cGMP synthesis in photoreceptors. Mammalian GCAPs share over 90% sequence similarity, consisting of 201–205 amino acids with three conserved Ca²⁺-binding sites.
The GUCA1A gene (also known as GCAP1) is expressed only in the retina. Mutations such as Tyr99Cys and Pro50Leu have been associated with autosomal dominant cone dystrophy and variable retinal phenotypes. These mutations may reduce protein stability and increase susceptibility to protease degradation, contributing to retinal cell death.
