Anaplastic Lymphoma Receptor Tyrosine Kinase (ALK) belongs to the insulin receptor superfamily and plays a crucial role in brain development. Mutations, rearrangements, and amplifications of the ALK gene are implicated in various tumors, including anaplastic large cell lymphoma, neuroblastoma, and non-small cell lung cancer.
Chromosomal rearrangement often produces a fusion gene combining ALK and nucleophosmin (NPM), resulting in an oncogenic NPM-ALK product. The ALK protein contains transmembrane and extracellular domains absent in the NPM-ALK fusion. ALK shares high sequence similarity with LTK and influences specific neuronal development.
