Product Specific Information

Description:
The IT2.2 monoclonal antibody reacts with human CD86 (~80 kDa), also known as B7-2. CD86 and CD80 are costimulatory molecules expressed on B cells, macrophages, and dendritic cells. CD86 interacts with CD28 and CTLA-4 on T cells, playing a key role in T-B cell crosstalk, T cell costimulation, and antibody production.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and validated for flow cytometry of normal human peripheral blood cells at 5 µL (0.5 µg) per test. A test is defined as staining a sample in 100 µL volume. Cell numbers may range from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass.

Usage Recommendations:

• Use Super Bright Complete Staining Buffer (Product # SB-4401) when combining multiple Super Bright dyes to reduce non-specific interactions.
• Protect from light due to photo-induced oxidation sensitivity.
• Fixed samples can be stored up to 3 days at 4°C using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333).

Target Information

CD86 is a ligand for CTLA4 and CD28, acting as a costimulatory molecule during antigen presentation. Insufficient co-stimulation can induce tolerance. CD86 is expressed by antigen-presenting cells and is involved in T-cell activation and immune regulation. Alternative splicing produces multiple isoforms. CD86 dysfunction is associated with gallbladder squamous cell carcinoma and myocarditis.
