Target Information
STK3 and STK4 are stress-activated, pro-apoptotic kinases that, following caspase cleavage, translocate to the nucleus and induce chromatin condensation and DNA fragmentation. They are core components of the Hippo signaling pathway, which regulates organ size and tumor suppression by controlling cell proliferation and apoptosis.
STK3/MST2 and STK4/MST1, in complex with SAV1, activate LATS1/2, which phosphorylate and inactivate YAP1 and WWTR1/TAZ, thereby preventing nuclear translocation and gene activation related to proliferation and migration.
These kinases are essential for repressing hepatocyte proliferation, preventing liver stem cell activation, and inhibiting tumor formation. They phosphorylate NKX2-1, NEK2, CROCC, CEP250, MOBKL1A/B, RASSF2, and regulate RAF1 activation and ESR1 transcriptional activity.
