Target Information
The ABL1 proto-oncogene encodes a cytoplasmic and nuclear protein tyrosine kinase involved in cell differentiation, division, adhesion, and stress response.
Activity of c-Abl protein is negatively regulated by its SH3 domain; deletion of this domain converts c-Abl into an oncogene.
CDC2-mediated phosphorylation regulates the DNA-binding activity of ABL1, linking it to cell cycle control.
In chronic myelogenous leukemia and some acute lymphoblastic leukemias, ABL1 undergoes a (9;22) translocation forming the Philadelphia chromosome, fusing c-Abl sequences with the Bcr gene.
The c-Abl oncogene was first identified as the transforming gene of Abelson murine leukemia virus (A-MuLV).
c-ABL may regulate DNA repair by activating proapoptotic pathways when damage is irreparable.
