Cyclin D1 (PRAD-1, bcl-1)은 세포 주기의 주요 조절 인자로, CDK4 및 CDK6과 결합하여 Rb 단백질을 인산화함으로써 세포 주기 진행을 조절합니다. Cyclin D1은 mantle cell lymphoma(MCL)을 포함한 다양한 인간 종양에서 과발현되는 원형암유전자(proto-oncogene)로 알려져 있습니다.
이 단백질은 G1 phase 동안 합성되며 성장인자 자극에 의해 CDK4 또는 CDK6과 복합체를 형성합니다. Cyclin D1-CDK 복합체는 Rb 단백질의 성장 억제 기능을 비활성화하고, CDK 억제제(p21^Cip1, p27^Kip1)의 작용을 조절합니다.
Cyclin D1은 성장인자 자극이 없을 때 불안정하며, 인산화에 의해 유비퀴틴 매개 분해를 거쳐 G1/S phase 정지에 관여합니다.
Cyclin D1은 세포주기 동안 단백질 양이 주기적으로 변하는 highly conserved cyclin family에 속하며, CDK kinase의 조절자로 기능합니다. Cyclin D1은 CDK4/CDK6과 복합체를 형성하여 G1/S 전환에 필수적인 역할을 하며, 종양억제 단백질 Rb와 상호작용합니다.
