Product Specific Information

  • Heat-mediated antigen retrieval using Tris/EDTA (pH 8) is recommended for paraffin sections.
  • Suggested positive control: Human tonsil.
  • Mouse anti-Human CD8 antibody (clone 4B11) recognizes the human CD8 cell surface antigen (~32 kDa glycoprotein), expressed by cytotoxic/suppressor T cells and weakly by NK cells.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family involved in signal transduction, T cell differentiation, and activation.
CD8 is predominantly expressed on T cells as a disulfide-linked heterodimer (CD8α/CD8β) functioning as a co-receptor for MHC-I molecules, while CD4 acts as a co-receptor for MHC-II.
CD8α chain is essential for MHC-I binding and is also expressed on subsets of T cells, NK cells, monocytes, and dendritic cells as homodimers.
Binding of MHC-I/peptide complexes triggers recruitment of Lck kinase, leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are crucial for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
