Target Information

Dihydropyridine receptor (DHPR) is a surface membrane protein critical for the excitation-contraction coupling of striated muscle. DHPR and the sarcoplasmic reticulum ryanodine receptor (RyR) are key components of intracellular junctions, converting membrane depolarization into Ca²⁺ release from internal stores. The alpha1-subunit of DHPR interacts with RyR via a cytoplasmic loop, and phosphorylation of DHPR alpha1 may modulate this interaction. Mutations in DHPR alpha1 cause excitation-contraction uncoupling, leading to muscular dysgenesis. Cells lacking RyR also exhibit reduced Ca²⁺ current density and lack excitation-contraction coupling.
