Activity
- Measured by its binding ability in a functional ELISA. Immobilized Mouse NOTCH1-His at 2 µg/mL (100 µL/well) can bind human DLL4-hFc, the EC50 of human DLL4-hFc is 15–90 ng/mL.
- Measured by the ability of the immobilized protein to enhance BMP2-induced alkaline phosphatase activity in C3H10T1/2 mouse embryonic fibroblast cells. The ED50 for this effect is typically 1–8 µg/mL in the presence of 500 ng/mL recombinant human BMP2.
Product Specific Information
- It is recommended that the protein be aliquoted for optimal storage.
- Add 500 µL of sterile water to the vial to prepare a stock solution of 0.25 mg/mL.

Target Information
Delta-like ligand 4 (DLL4), also known as Delta-4, is a homologue of the Drosophila delta protein and functions as a transmembrane ligand for the Notch1 receptor. DLL4 is expressed in the vasculature and plays a critical role in vascular development. It is induced by VEGF as a negative feedback regulator to control angiogenic sprouting and promote the formation of a differentiated vascular network. DLL4 is strongly expressed in tumor vessels of clear-cell renal tumors and bladder cancer, and inhibition of DLL4 leads to increased vascular proliferation but defective maturation, resulting in reduced tumor growth without apparent toxicity.
