Product Specific Information
This clone recognizes the 60 kDa subunit 1 (B-subunit, the product of the VMA2 or VAT2 gene) of S. cerevisiae.
By Western blot, it can be used to identify organelles during subcellular fractionation of yeast membranes.
Target Information
Vacuolar-type H⁺-ATPase (V-ATPase) is a multisubunit enzyme responsible for acidification of eukaryotic intracellular organelles.
V-ATPases pump protons against an electrochemical gradient, while F-ATPases reverse the process to synthesize ATP.
The enzyme consists of:
- Peripheral V1 domain (ATP hydrolysis)
- Integral V0 domain (proton translocation)
The V1 domain has nine subunits (A–H), and the V0 domain has five subunits (a, d, c, c′, c″).
Like F-ATPase, V-ATPase likely operates through a rotary mechanism.
The V-ATPase V1 B subunit exists as two isoforms:
- B1 isoform: functions in proton secretion in the inner ear, maintaining endolymph pH and auditory function.
Mutations in this gene cause distal renal tubular acidosis with sensorineural deafness. - B2 isoform: expressed in kidney and osteoclasts (only B isoform in osteoclasts).
