Specificity Comments:
Cytotoxic T lymphocyte (CTL)-mediated cytotoxicity plays a key role in immune surveillance against virus-infected or transformed cells. Two mechanisms are known: perforin-based and FAS-based. The FAS-based mechanism involves FAS (Apo-1) and its ligand FAS-L. FAS is a cell surface glycoprotein belonging to the TNF receptor family, expressed on various lymphoid cell lines that undergo apoptosis upon antibody treatment. These findings suggest that FAS–FASL interactions mediate targeted cell death and are critical in CTL-mediated cytotoxicity.

Target Information

CD178 (Fas ligand, FasL) is a type-II membrane protein with its N-terminus in the cytoplasm and C-terminal region extending extracellularly. Its receptor, FasR, is a type-I membrane protein belonging to the TNF and NGF receptor families. CD178 induces apoptosis upon binding to FasR and exists in both membrane-bound (45 kD) and soluble (26 kD) forms. The soluble form is released by metalloproteinase cleavage.
Binding of CD178 to Fas triggers receptor oligomerization and apoptotic signaling. CD178 is predominantly expressed in activated T lymphocytes and NK cells, and also in immune-privileged tissues such as testis and eye. Expression is reported in thymus, liver, ovary, lung, heart, and kidney.
CD178-mediated apoptosis is involved in antiviral immune responses and immune regulation. The Fas/CD178 system is implicated in diseases such as AIDS, hepatitis, cancer, atherosclerosis, and inflammatory conditions.
