Product Specific Information

The HIB19 monoclonal antibody reacts with human CD19, a 95 kDa transmembrane glycoprotein expressed by B cells during all stages of development except terminally differentiated plasma cells. Follicular dendritic cells also express CD19. CD19 forms a multimolecular complex with CD21, CD81, Leu13, and MHC class II, associating with BCR to induce tyrosine phosphorylation, calcium flux, and proliferation of B cells.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (1.0 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample to antibody cocktails before combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample.

Based on Phiton™ technology utilizing novel nucleic acid dye structures engineered for optimized fluorescent signatures, minimizing spillover and spread.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on 100% of peripheral B cells and some myeloid leukemia cells. CD19 acts as a signal-amplifying co-receptor for the B cell receptor (BCR) and regulates B cell proliferation and differentiation. Mutations lead to hypogammaglobulinemia, while overexpression causes B cell hyperactivity.
