Target Information
MDM2는 p53의 유비퀴틴 리가아제로서 p53 안정성 조절의 중심적인 역할을 수행합니다. MDM2는 염색체 12q에 위치하며, Akt에 의해 Ser166 및 Ser186에서 인산화될 때 p300과의 상호작용이 증가하여 MDM2 매개 p53 유비퀴틴화 및 분해를 촉진합니다. 또한 인산화된 MDM2는 p19ARF와의 결합을 차단하여 p53 분해를 증가시킵니다. MDM2는 p53 기능을 음성 조절하며, 과발현 시 p53의 종양 억제 기능을 감소시킬 수 있습니다. 이 단백질은 또한 E3 유비퀴틴 리가아제 활성을 통해 p53을 분해합니다. MDM2는 세포자멸사(apoptosis)와 세포주기 조절에도 중요한 역할을 하며, 다양한 인간 암(예: 연조직암, 유방암)에서 과발현됩니다. p53 외에도, MDM2는 retinoblastoma 1 및 ribosomal protein L5 등 여러 단백질과 상호작용하여 세포주기, 세포사멸, 종양형성 과정에 관여합니다. 40개 이상의 다양한 스플라이싱 변이체가 종양 및 정상 조직에서 확인되었습니다.
