Target Information

IL-33R, also known as ST2, is a receptor for IL-33 and a member of the IL-1 receptor family. Its extracellular domain contains three Ig-like domains that interact with the co-receptor IL-1RAcP and ligand IL-33. Upon IL-33 binding, the IL-33R complex recruits adaptor proteins such as MyD88, IRAK1, IRAK4, and TRAF6, leading to activation of NF-κB and MAPK pathways. This signaling induces the release of cytokines including IL-5, IL-6, IL-8, and IL-13.
Two main isoforms, ST2L (membrane-bound) and ST2S (soluble), are generated by alternative splicing. The soluble form can act as a decoy receptor, modulating IL-33 signaling. IL-33R is primarily expressed by mast cells, basophils, and eosinophils, and in mice, also by ILC2 and Th2 cells. IL-33R signaling is associated with immune responses such as allergy, arthritis, atherosclerosis, and sepsis.
