Product Specific Information

The eBio1D4B monoclonal antibody reacts with mouse CD107a (LAMP-1), a type I lysosomal membrane protein that is highly glycosylated. CD107a is constitutively expressed in late endosomes and lysosomes of all cells and transiently expressed on the surface of degranulating cytolytic T cells. It is involved in various cellular functions including cancer metastasis and serves as a marker for lysosomal storage disorders.

Target Information

LAMP1 (CD107a), together with LAMP2, is a major lysosomal membrane constituent. Approximately 1–2% of CD107a is also found on the plasma membrane. LAMP1 is a heavily glycosylated type I integral membrane protein containing a signal peptide, 18 N-linked glycosylation sites, a single membrane-spanning segment, and a short cytosolic tail.

LAMP proteins are essential for lysosome biogenesis and fusion with phagosomes. Upon cell activation, LAMP1 translocates to the plasma membrane via a carboxyl-terminal tyrosine-based motif (YXXI). Mutations affecting this motif disrupt lysosome localization.

Cell surface LAMP1 and LAMP2 promote adhesion of PBMCs to vascular endothelium, suggesting involvement in inflammation sites. Increased LAMP1 immunoreactivity is observed in neurons and glial cells near senile plaques in Alzheimer’s disease and in thymic epithelial cells during acute involution. LAMP1 is a reliable marker of mast cell activation.
