Product Specific Information

This 4D10.1 monoclonal antibody recognizes human Syk, a 72-kDa member of the Syk/ZAP-70 family of non-receptor protein tyrosine kinases.
Syk is highly expressed in B cells, with lower expression in immature thymocytes, mast cells, and platelets. It can also be detected in fibroblasts, epithelial cells, hepatocytes, endothelial cells, and neuronal cells.
Syk plays an essential role in lymphocyte development and signaling cascades downstream of B and T cell antigen receptors. Upon recruitment to ITAMs, Syk is activated by phosphorylation and subsequently phosphorylates proteins such as phospholipase C gamma and BLNK/SLP-65.
Abnormal Syk expression has been linked to tumor cell migration and invasion in several cancers.

Applications Reported: Flow cytometric analysis and Western blotting.
Applications Tested: Immunoblotting of reduced Ramos cell lysate at ≤5 µg/mL.
Careful titration is recommended for optimal assay performance.

Spleen tyrosine kinase (SYK) and ZAP70 are cytoplasmic tyrosine kinases considered potential drug targets for allergic and autoimmune diseases.
SYK translocates to the plasma membrane upon BCR or high-affinity IgE receptor (FcεRI) activation, phosphorylating adaptor proteins and initiating downstream signaling such as calcium mobilization, cytoskeleton remodeling, and gene transcription.
SYK binds receptor assemblies via SH2 domain interactions with ITAM motifs phosphorylated by Src-family kinases, which also activate SYK via phosphorylation of its activation loop.
