Target Information
NCSTN (Nicastrin) encodes a type I transmembrane glycoprotein that is part of the multimeric gamma-secretase complex. This protein cleaves integral membrane proteins such as Notch receptors and beta-amyloid precursor protein and may act as a stabilizing cofactor for gamma-secretase complex assembly.
Cleavage of beta-amyloid precursor protein generates amyloid beta peptide, the main component of neuritic plaques characteristic of Alzheimer’s disease. However, the exact role of Nicastrin in Alzheimer’s disease remains uncertain.
Mutations in NCSTN are associated with familial acne inversa, and a pseudogene is located on chromosome 21.
