Target Information
TSC2 (Tuberin, Tuberous sclerosis complex, TSC Complex Subunit 2)은 종양 억제 인자로 알려져 있으며, 세포 성장 정지 조절 및 스테로이드 수용체 매개 전사 조절에 관여할 수 있습니다. TSC2는 세포질 복합체에서 hamartin과 결합하여 샤페론 역할을 할 수 있으며, Ser939 및 Thr1462의 인산화는 hamartin과의 상호작용을 조절합니다. 이러한 인산화는 phosphoinositide 3-kinase/Akt 경로에 의해 성장 인자에 의해 자극됩니다.
TSC2는 세포 성장 정지 조절, 소포 수송 조절, 스테로이드 수용체 매개 전사 조절에 중요한 역할을 하며, TSC1과의 상호작용은 소포 도킹을 촉진할 수 있습니다. 또한 TSC2는 RAP1A 및 RAB5 단백질의 GTPase 활성을 촉진하여 세포 성장 조절에 관여합니다. TSC2 유전자 변이는 RAP1A의 지속적 활성화를 유도하며, 림프관평활근종증(lymphangioleiomyomatosis) 및 TSC2 angiomyolipoma와 같은 질환과 관련이 있습니다. 대체 스플라이싱을 통해 세 가지 아이소폼의 전사 변이가 보고되었습니다.
