Target Information

RIPK2 is a serine/threonine kinase involved in cell activation, proliferation, differentiation, and apoptosis. Upon stimulation by bacterial peptidoglycans, NOD1 and NOD2 are activated, oligomerize, and recruit RIPK2 through CARD–CARD domains. RIPK2 contributes to the tyrosine phosphorylation of ARHGEF2 via Src kinase, leading to NF-κB activation by NOD2. Once recruited, RIPK2 autophosphorylates and undergoes Lys-63–linked polyubiquitination by E3 ubiquitin ligases XIAP, BIRC2, and BIRC3. This modification facilitates recruitment of MAP3K7/TAK1 to IKBKG/NEMO, inducing further Lys-63–linked polyubiquitination and activation of IKBKB/IKKB. Consequently, NF-κB is released, translocates to the nucleus, and activates transcription of numerous genes involved in immune response, growth control, and apoptosis protection. RIPK2 also promotes BCL10 phosphorylation and NF-κB activation during T-cell receptor engagement.
