Product Specific Information

The 6B10.2 monoclonal antibody reacts with human CD247 (CD3 zeta), also known as the invariant zeta chain.
The CD3 complex consists of gamma, delta, and epsilon monomeric chains and the invariant zeta chain, forming homo- or heterodimers with the epsilon subunit. Together with the TCR, these chains compose the TCR/CD3 complex, expressed on immature and mature thymocytes, T lymphocytes, and NK cells.

Signal transduction during T cell activation occurs via tyrosine phosphorylation of ITAMs on the zeta chain.
The 6B10.2 antibody recognizes a transmembrane epitope of CD3 zeta, thus requiring intracellular staining for flow cytometric analysis. It recognizes the zeta chain as a homodimer, not as a monomer.

Target Information

The CD3z complex is essential for transducing antigen-recognition signals in T cells and regulating TCR surface expression.
It comprises CD3 gamma, delta, epsilon, and zeta subunits—members of the immunoglobulin superfamily encoded on human chromosome 11. The cytoplasmic tails of CD3 subunits associate with signal transduction molecules via double tyrosine-based motifs.

CD3 is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum. It is not expressed on B or NK cells.
Decreased T lymphocyte percentages may be associated with autoimmune diseases.
Diseases linked to CD3z include Immunodeficiency 25 and T-B+ Severe Combined Immunodeficiency due to CD3δ/ε/ζ defects.
