The Human IL-2 Simplex ProcartaPlex Kit measures IL-2 protein and is designed to be combined with other Simplex kits to create custom multiplex panels utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits (i.e., when not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is required per assay plate regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on the sandwich ELISA principle, using two highly specific antibodies binding to different epitopes of a protein to quantitate multiple targets simultaneously using a Luminex instrument. These assays require as little as 25 μL of plasma or serum, or 50 μL of cell culture supernatant, and deliver analyzed results in approximately four hours.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single sample. Luminex beads are internally dyed with red and infrared fluorophores, creating unique spectral signatures identified by Luminex xMAP detection systems (e.g., Luminex 200, FLEXMAP 3D, MAGPIX).

Similar to a sandwich ELISA, each bead is labeled with antibodies specific for a single target protein. Bound proteins are identified with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The conjugation of antibodies to distinct beads enables multiplexed analysis in a single well.

Unlike ELISA, the capture antibody in a ProcartaPlex assay is conjugated to a bead rather than adsorbed to a microplate well, allowing reagents to remain free-floating in solution. Detection is performed using dual-laser instruments (e.g., Luminex 200), where one laser identifies bead spectral signatures and the other quantifies RPE fluorescence proportional to protein concentration.

ProcartaPlex multiplex assays can profile more targets using significantly less sample in the same time required for a traditional ELISA. Multiplex panels are available across six species (human, mouse, rat, nonhuman primate, porcine, and canine).
