Product Specific Information

The eBioH4A3 monoclonal antibody reacts with human CD107a (LAMP-1), a glycosylated protein (~110 kDa) mainly expressed in lysosomal/endosomal membranes. It is transiently expressed on the surface of degranulating cytolytic T cells and upregulated on activated platelets and certain cancer cells.

Applications Reported:

  • Intracellular staining followed by flow cytometric analysis
  • Surface staining in flow cytometric-based degranulation assays

Applications Tested:
Pre-titrated and tested by intracellular staining and flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test.

Light Sensitivity:
Protect vial and stained samples from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-822-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Excitation/Emission:
488–561 nm / 667 nm (Blue, Green, Yellow-Green lasers)

Filtration:
0.2 µm post-manufacturing filtered.


Target Information

LAMP1 (CD107a) is a lysosome-associated membrane protein involved in lysosome biogenesis and fusion with phagosomes. It is a type I integral membrane protein with multiple N-linked glycosylation sites and a short cytosolic tail. Upon activation, LAMP1 translocates to the plasma membrane via a tyrosine-based motif (YXXI). Surface LAMP1 promotes adhesion of PBMCs to vascular endothelium, potentially contributing to inflammatory responses. Increased LAMP1 expression is observed in neurons and glial cells around senile plaques in Alzheimer’s Disease, and serves as a marker of mast cell activation.
