Product Specific Information

This TXRX10 monoclonal antibody reacts with human and mouse TOX (Thymocyte selection-associated HMG box protein). TOX is a DNA-binding nuclear factor expressed in thymocytes, T lymphocytes, NK cells, and lymphoid tissue-inducer (LTi) cells. Expression is upregulated during beta-selection and positive selection in thymocytes.
Studies using Tox knockout mice have shown TOX is essential for CD4⁺ T cell development, particularly in the CD4loCD8lo → CD4⁺CD8lo transition. It also plays roles in NK and LTi cell development. TOX expression is induced by calcineurin-mediated TCR signaling during positive selection.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Intracellular flow cytometry of mouse thymocytes using Foxp3 Buffers. Recommended ≤ 0.5 µg/test (100 µL final volume).

Target Information

TOX (Thymocyte selection-associated high mobility group box protein) is a member of the HMG box family of DNA-binding proteins involved in T-cell development regulation. It is a 526 amino acid nuclear protein, upregulated by pre-TCR and TCR activation in immature thymocytes.
TOX-mediated positive selection is associated with RUNX3 upregulation and is calcineurin dependent. The TOX-dependent CD4⁺CD8 transition is required for development of class II MHC-specific T cells.
