Target Information

Mitochondrial transcription factor A (TFAM) is a nuclear-encoded protein that translocates to mitochondria, where it plays a crucial role in maintaining and expressing mitochondrial DNA (mtDNA). TFAM regulates mtDNA replication and transcription, ensuring proper mitochondrial function. It contains an LC3 interacting region (LIR) motif, enabling binding to autophagy proteins such as LC3, which supports autolysosomal degradation of leaked mtDNA and limits inflammation.

TFAM levels vary across tissues, influencing mitochondrial function. High TFAM levels can repress mtDNA expression and affect oxidative phosphorylation (OXPHOS) in some tissues, while in others, such as the heart, increased mtDNA copy number maintains balanced TFAM-to-mtDNA ratios. Disruptions in TFAM are associated with mitochondrial dysfunction, neurodegeneration, and other mtDNA-related diseases.
